Carbon Monoxide Inhibition of Azotobacter in Microrespiration Experiments.

چکیده
Extremely small quantities of carbon monoxide in air inhibit nitrogen fixation by red clover plants inoculated with Rhizobium trifolii (Lind and Wilson, 1941). In preliminary trials with Azotobacter vinelandii Wyss (1941) found a similar inhibition at carbon monoxide concentrations about 10 times those required for the symbiotic system. We have made a detailed study of the inhibition of this azotobacter using several sources of combined nitrogen to determine specificity for the nitrogen fixation process. Two methods have been used, each of which possesses certain advantages: a macro method which periodically measures directly the nitrogen fixed and a micro method which indirectly estimates nitrogen fixation by determining the increase in rate of respiration with time. The macro method is relatively longtime, 30-40 hr in comparison with the 4-7 hr required in the respiration studies. The results of the macro experiments have been described by Lind and Wilson (1942); in this paper the micro-respiration data are discussed.

Action of Inhibitors on Hydrogenase in Azotobacter
The inhibitors usually associated with the activity of the cytochrome oxidase system-cyanide and carbon monoxide-are also effective in reducing the oxidation of H(2) by intact cells of Azotobacter vinelandii. The hydrogenase system is more sensitive to CO than is the respiratory system. Oxidation of a carbon source and of hydrogen by Azotobacter cells is inhibited in a quantitatively different ...
متن کاملInteractions among substrates and inhibitors of nitrogenase.
Examination of interactions among various substrates and inhibitors reacting with a partially purified nitrogenase from Azotobacter vinelandii has shown that: nitrous oxide is competitive with N2; carbon monixide and acetylene are noncompetitive with N2; carbon monoxide, cyanide, and nitrous oxide are noncompetitive with acetylene, whereas N2 is competitive with acetylene; carbon monoxide is no...
متن کامل93. Mechanism of Biological Nitrogen Fixation
A PREVIOuS report [Wyss & Wilson, 1941] demonstrated that B2 inhibits fixation of N2 by Azotobacter. Because of the apparent uniqueness of this type of inhibition, such a demonstration furnishes strong evidence that in the symbiotic and free-living N2 fixation processes the mechanism of fixation is very similar if not identical. Further studies described in this paper provide additional support...
متن کاملThe in vivo hydrocarbon formation by vanadium nitrogenase follows a secondary metabolic pathway
The vanadium (V)-nitrogenase of Azotobacter vinelandii catalyses the in vitro conversion of carbon monoxide (CO) to hydrocarbons. Here we show that an A. vinelandii strain expressing the V-nitrogenase is capable of in vivo reduction of CO to ethylene (C2H4), ethane (C2H6) and propane (C3H8). Moreover, we demonstrate that CO is not used as a carbon source for cell growth, being instead reduced t...
متن کاملEnzymatic oxidation of tetramethyl-p-phenylenediamine and p-phenylenediamine by the electron transport particulate fraction of Azotobacter vinelandii.
The ability of the electron transport particulate fraction of Azotobacter vinelandii strain O to oxidize tetramethyl-p-phenylenediamine (TMPD) and p-phenylenediamine (PPD) was examined in detail. The highest specific activity for TMPD and PPD oxidation concentrated in the A. vinelandii O R(3) fraction. The A. vinelandii O R(3) fraction was used to develop a standard manometric assay which gave ...
